According to the result of spread-plate method on nutrient agar plates, a number of colonies in C-1 plates were observed as follows: 13 and 11 colonies (average 12 colonies).
In addition, 6 and 5 colonies were observed in H-2 and H-1 plates, respectively. However, there were 0 colonies observed for 2nd trial of both H-2 and H-1 plates.
This indicates that some sort of error has occurred in the first place, most likely due to spreading or contamination errors. As a result, our group was instructed from TA to consider 6 colonies are observed in H-1 plate, rather than H-2 plate. Hence, the average number of colonies in H-1 plate was calculated to be 5.5 colonies.
Finally, these counts were calculated in log CFU/g unit: 2.381 for C-1 plate and 2.041 for H-1 plate.
In contrast, the result of petri-film method showed that 5 and 13 colonies (average 9 colonies) were detected in C-1 dilution, whereas 0 colonies were detected in H-0 dilution.
The fact that no colonies detected in H-0 petri-film further supports that spreading
