원리를 공부한다. 이 방법을 이용하여 미지 단백질의 정량을 분석한다.

5. Theory : Gel Filtration Chromatography
GEL Filtration Chromatography
Some source refer to this as molecular sleve or gel filtration chromatography. Regardless of which mane is applied, the fundamental principle is simple.
Exclusion media may be prepared from organic or inorganic materials, fabricated in a way that produces multitudinous fine channels or pores thru the mass of the material. With organic media, the pores are produed by cross linking of the polymer. W/ inorganic materials the pores are produced by an etching or solution process that removes part of the mass : this is the method by which porous glass beads are produced. When a slurry of an exclusion medium in buffer is poured as a column bed, the total coumn vol, Vt., may be regarded as containing three compartments :
Vt = Vo + Vm + Vi
Where Void the vol. Xternal to the medium, Vm is the vol. Of the gel itself, and Vi is the Vol. Included within the gel. A solute spcecious will distribute between Vo and Vi w/ a distribution coefficient, Kd. We can at once write that
Ve = V0 + KdVi
